On the stickie it says that I have to download the game and unzip but I cant even get to that point because when I attempt to unzip thats when it says it doesnt support full screen mode. Should I be doing something in dosbox before I unzip?

-> Unzipping is done in Windows.
-> Maybe it would be a good idea to state about WHAT Duke you are talking?
There are 3 of them:
DN1
DN2
DN3D
And from what site you've downloaded it (This forum covers TWO sites)

[EDIT]
Should you need a unzipping program:
http://www.7-zip.org/
Freeware, works fine in Vista, the one you get with Vista is too basic anyway.

I am having a similar problem with Alley Cat. I can extract the file, but get this message when I try and open it. I get a pop-up box for 16 bit MS-DOS subsystem saying "This system does not support full-screen mode. Select close to terminate this application." You can also choose to "Ignore" but then it says the program has to close because of problems. I have an HP computer with Vista. Any ideas?

The only way to get that game running in Vista is dosbox.
There is a sticky especially for that:
